Do I need to upload the MDB file every load?

No, you don’t. The MDB file is automatically selected and populated based on the dictionary type.

You need to upload the MDB file only when there is a new MDB released or if a custom MDB needs to be used. To do so, use the Upload New Configuration File option from the Dictionary Loader screen.
